Ocean-caught large yellow croaker (Larimichthys crocea) constitutes a crucial gene pool for the breeding of this species, yet these fish experience problematic survival rates in captivity and are unsuitable for breeding programs. In contrast to the utilization of wild-caught croakers, a proposal has been made for germ cell transplantation. L. crocea specimens will be the donors, and yellow drum (Nibea albiflora) will be the recipients. The identification of L. crocea and N. albiflora germ cells forms a critical foundation for the creation of a germ cell transplantation protocol for these fish. In N. albiflora, the 3' untranslated regions (UTRs) of the vasa, dnd, and nanos2 genes were cloned through the rapid amplification of cDNA ends (RACE) method, and then the obtained sequences were subjected to alignment and analysis in comparison to L. crocea and N. albiflora. Based on the disparities in gene sequences, we crafted species-unique primers and probes that were applied to RT-PCR and in situ hybridization methods. Through RT-PCR, species-specific primers selectively amplified DNA from the gonads of each species, thus solidifying the efficacy of our six primer sets for identifying germ cells in L. crocea and N. albiflora. Utilizing in situ hybridization, we observed that the Lcvasa and Nadnd probes exhibited strong species-specific targeting, while the probes for Navasa and Lcdnd demonstrated reduced specificity. In situ hybridization, leveraging Lcvasa and Nadnd, permitted the visualization of germ cells in the two studied species. Thanks to these species-specific primers and probes, we can confidently distinguish the germ cells of L. crocea and N. albiflora, thereby creating a dependable strategy for recognizing germ cells post-transplantation, when L. crocea and N. albiflora serve as donor and recipient, respectively.

Ascomycota and Basidiomycota constituted the predominant components of the soil fungal community, achieving a relative abundance surpassing 90%. Fungal diversity in the topsoil exhibited no significant altitudinal variation, in contrast to the subsoil's diversity, which decreased with increasing altitude. A greater degree of fungal diversity was observed within the topsoil. The level of soil fungal diversity was considerably impacted by the altitude.
